SECONDARY ANTIBODIES FOR WESTERN BLOTTING

READY-TO-USE

STABLE AT ROOM TEMPERATURE

Secondary antibodies conjugated to HRP (horseradish peroxidase) or AP (alkaline phosphatase), stable at ambient temperature for several weeks, are now available. They can be stored at 2-8 oC for even longer life expectancy (18 to 24 months). In addition, they need no further dilution or aliquoting, since they are formulated in a ready-to-use solution. The conjugates are suitable for both chromogenic staining and chemiluminescence detection protocols using either PVDF or nitrocellulose membranes.

Advantages over lyophilized or concentrated forms:

  • Excellent reproducibility.
  • No need to dissolve and/or dilute.
  • No need to aliquot and freeze.
  • No need to worry about shipping and storage conditions.
  • Maximized life expectancy.
  • Customized quantities of the same batch are available upon request.

The thermostable and ready-to-use secondary antibody conjugates offered by ALTER CHEM for Western Blotting produce more intense signal of the specific bands and lower background as shown in the figure below.

A human cell line extract has been analyzed by SDS-PAGE and transferred to membrane. The blot has been incubated with anti-PARP antibody (rabbit polyclonal). Then incubated with ALTER CHEM's thermostable & ready-to-use anti-rabbit IgG-HRP conjugate (lane 1) or with an anti-rabbit IgG-HRP conjugate from Company B (lane 2) (according to the manufacturer's instructions). Finally, the blot was incubated with chemiluminescence reagent and image acquired with a phosphorimager. The lane 3 represents electrophoretic analysis and blotting of a mixture of the human cell line extract and a mouse epithelial cell line extract, which was analyzed as previously described, omitting the incubation with primary (anti-PARP) antibody
